Overview
MRG003 + toripalimab is an investigational combination therapy consisting of becotatug vedotin (MRG003), an antibody-drug conjugate (ADC) targeting the epidermal growth factor receptor (EGFR), and toripalimab, a monoclonal antibody targeting programmed cell death protein 1 (PD-1). MRG003 utilizes a humanized anti-EGFR antibody conjugated to the microtubule-disrupting agent monomethyl auristatin E (MMAE) via a valine-citrulline cleavable linker. Toripalimab acts as an immune checkpoint inhibitor to enhance the anti-tumor immune response. This combination is being evaluated in Phase 2 clinical trials as a neoadjuvant treatment for patients with PD-L1-positive, resectable, locally advanced head and neck squamous cell carcinoma (LA-HNSCC), aiming to improve pathological response rates and surgical outcomes.
