Belantamab mafodotin + lenalidomide is a combination therapy being investigated for the treatment of multiple myeloma. Belantamab mafodotin (Blenrep) is an antibody-drug conjugate (ADC) developed by GSK that targets B-cell maturation antigen (BCMA). It consists of a humanized anti-BCMA monoclonal antibody conjugated to the microtubule inhibitor monomethyl auristatin F (MMAF) via a non-cleavable maleimidocaproyl linker. Lenalidomide (Revlimid) is an oral immunomodulatory imide drug (IMiD) that binds to cereblon, part of the E3 ubiquitin ligase complex, leading to the degradation of Ikaros and Aiolos transcription factors. This combination is specifically being evaluated by Roswell Park Cancer Institute in a Phase 2 trial as consolidation therapy for patients with multiple myeloma who remain minimal residual disease (MRD) positive after autologous stem cell transplantation (ASCT).
