Overview

Belimumab + cyclosporine is an investigational combination therapy being evaluated for the treatment of corticosteroid-refractory primary immune thrombocytopenia (ITP). Belimumab is a human monoclonal antibody that targets and inhibits B-lymphocyte stimulator (BLyS), also known as BAFF, which is a cytokine essential for B-cell survival and maturation. By inhibiting BLyS, belimumab reduces the survival of autoreactive B-cells and the production of autoantibodies. Cyclosporine is a small molecule calcineurin inhibitor that suppresses T-cell-mediated immune responses by inhibiting the transcription of interleukin-2 and other cytokines necessary for T-cell activation. The combination aims to provide a synergistic therapeutic effect by simultaneously targeting both B-cell and T-cell pathways involved in the autoimmune destruction of platelets in ITP. This regimen is currently being studied in clinical trials led by researchers at The Second Affiliated Hospital of Kunming Medical University.
