Belinostat + carboplatin + paclitaxel is a combination chemotherapy regimen investigated primarily for the treatment of recurrent or platinum-resistant epithelial ovarian cancer and other solid tumors. Belinostat is a histone deacetylase (HDAC) inhibitor that modulates gene expression by inhibiting HDAC enzymes, leading to cell cycle arrest and apoptosis in cancer cells. Carboplatin is a platinum-based chemotherapeutic agent that causes DNA crosslinking and subsequent apoptosis. Paclitaxel stabilizes microtubules and inhibits their disassembly, disrupting mitosis and promoting cell death. The combination has been evaluated in phase I/II clinical trials; it was generally well tolerated with manageable toxicity profiles but did not show significant improvement in progression-free survival compared to standard regimens. The regimen has demonstrated some clinical benefit in heavily pretreated patients with ovarian cancer[1][2][3].
