Belinostat + doxorubicin is a combination of **belinostat** (a histone deacetylase inhibitor, HDACi) and **doxorubicin** (an anthracycline cytotoxic agent) used in investigational settings, primarily for advanced or metastatic soft tissue sarcoma (STS). Belinostat acts by inhibiting histone deacetylase enzymes, resulting in increased acetylation of histone proteins, altered gene expression, cell cycle arrest, and apoptosis in tumor cells. Doxorubicin intercalates into DNA and inhibits topoisomerase II, leading to DNA damage and cell death. This combination was evaluated in Phase I/II trials, where belinostat was administered intravenously over 5 days and doxorubicin was administered intravenously on day 5 of each 3-week cycle. The regimen showed moderate clinical activity and was generally well-tolerated, with main toxicities including fatigue, nausea, alopecia, vomiting, constipation, and neutropenia[1][2][3][4][6].
