Overview
**Belinostat + idarubicin** is a combination of two chemotherapeutic agents: belinostat, a histone deacetylase (HDAC) inhibitor, and idarubicin, an anthracycline antibiotic. This combination has been studied primarily in the context of acute myeloid leukemia (AML) and acute promyelocytic leukemia (APL). Belinostat induces cell cycle arrest and apoptosis by inhibiting HDAC-mediated epigenetic regulation, while idarubicin intercalates DNA and inhibits topoisomerase II, leading to inhibition of DNA replication and repair. Preclinical studies suggest that the combination can synergistically increase apoptosis and impair metabolic activity in leukemic cells, and early clinical trials have demonstrated anti-leukemic effects in AML patients[1][3].
