Belinostat + ribociclib is an investigational combination therapy consisting of the histone deacetylase (HDAC) inhibitor belinostat and the cyclin-dependent kinase 4/6 (CDK4/6) inhibitor ribociclib. This combination is primarily being evaluated for the treatment of metastatic triple-negative breast cancer (TNBC) and recurrent ovarian cancer. The rationale for the combination is based on preclinical evidence suggesting that HDAC inhibition can sensitize tumor cells to CDK4/6 inhibitors by modulating the retinoblastoma (Rb) protein pathway and increasing the expression of cell cycle inhibitors like p21. Belinostat is currently FDA-approved for peripheral T-cell lymphoma, while ribociclib is approved for HR+/HER2- breast cancer. The combination is being studied in the Phase I/Ib CHARGE trial (NCT04315233) sponsored by the University of Utah.
