**Belrestotug + iberdomide** is an investigational combination therapy composed of belrestotug, a human IgG1 monoclonal antibody targeting TIGIT (T cell immunoglobulin and ITIM domain), and iberdomide, a cereblon E3 ligase modulator (CELMoD agent). Belrestotug acts as an immune checkpoint inhibitor, aiming to enhance antitumor immune responses by blocking TIGIT, while iberdomide promotes immune system activation and tumor cell death by modulating cereblon and downstream substrates involved in myeloma and other hematologic malignancies. Belrestotug has primarily been developed for use in solid tumors, notably non-small cell lung cancer (NSCLC), but clinical trials for the combination specifically with iberdomide are currently unreported; both agents have separately been explored in combination therapies. Iberdomide is under investigation mainly for multiple myeloma.
