Drug intelligence / Profile preview
**belumosudil + azithromycin** is a combination of two drugs: - **Belumosudil** is a selective inhibitor of rho-associated coiled-coil–containing protein kinase-2 (ROCK2), designed to modulate immune function and fibrosis by inhibiting ROCK2 signaling, thereby reducing pro-inflammatory cytokine production and fibrosis through regulation of the Th17 and Tfh pathways and restoring immune homeostasis[2][4]. It is approved for chronic graft-versus-host disease (cGVHD) and under investigation for lung transplantation rejection and bronchiolitis obliterans syndrome (BOS)[1][3][4]. - **Azithromycin** is a macrolide antibiotic that inhibits bacterial protein synthesis by binding to the 50S ribosomal subunit. It is widely used for respiratory, skin, and other infections, and has additional anti-inflammatory properties used in chronic lung transplant settings[1][3]. This combination is specifically under investigation as part of standard immunosuppression regimens for patients with progressive lung allograft dysfunction (especially CLAD/BOS), and in trials for lung transplant rejection[1][3].
