Belzutifan + nivolumab is an **investigational combination** therapy currently being evaluated primarily for the treatment of advanced or metastatic renal cell carcinoma. **Belzutifan** is a selective small molecule inhibitor of **hypoxia-inducible factor 2 alpha (HIF-2α)**, a transcription factor implicated in tumorigenesis and tumor progression in clear cell renal cell carcinoma (ccRCC) and certain tumors associated with von Hippel-Lindau (VHL) disease. **Nivolumab** is a **monoclonal antibody** that blocks **programmed death-1 (PD-1)**, a checkpoint protein on immune cells, thereby enhancing antitumor immune responses. This combination aims to simultaneously inhibit hypoxia-induced tumor growth and overcome immune evasion, potentially offering improved efficacy. The regimen is under investigation (not yet approved as a fixed combination) in phase 1/2 studies for various solid tumors, most notably ccRCC.
