The combination of **bemarituzumab** (a humanized monoclonal antibody selective for fibroblast growth factor receptor 2b, FGFR2b) with **mFOLFOX6** (a standard chemotherapy regimen consisting of oxaliplatin, leucovorin, and 5-fluorouracil) is an investigational therapy for first-line treatment of unresectable locally advanced or metastatic gastric or gastroesophageal junction cancer (GC/GEJC) in patients with tumors that overexpress FGFR2b and are negative for HER2. Bemarituzumab inhibits FGFR2b, thereby suppressing tumor proliferation and promoting antibody-dependent cellular cytotoxicity. mFOLFOX6 acts as a cytotoxic chemotherapy regimen. The combination has shown clinically meaningful improvement in progression-free and overall survival compared to chemotherapy alone in phase 2 and phase 3 clinical trials for this patient population[1][2][3][4][5].
