**Bemarituzumab + nivolumab** is an investigational combination therapy for advanced gastric and gastroesophageal junction cancer with FGFR2b overexpression. Bemarituzumab is a first-in-class humanized monoclonal antibody selective for **fibroblast growth factor receptor 2b (FGFR2b)**, inhibiting its signaling to reduce tumor cell proliferation and enhance antibody-dependent cellular cytotoxicity. Nivolumab is a human IgG4 monoclonal antibody checkpoint inhibitor targeting **programmed cell death protein 1 (PD-1)**, enhancing anti-tumor immune response. Preclinical data and early-phase trials suggest bemarituzumab may sensitize tumors to anti-PD1 therapy, providing rationale for the combination. The regimen is currently under phase 1b and phase 3 investigation, primarily as first-line treatment for FGFR2b-overexpressed, HER2-negative, unresectable or metastatic gastric and GEJ adenocarcinoma[2][3][5][7].
