**Bempegaldesleukin + nivolumab** is a combination therapy consisting of bempegaldesleukin, a pegylated interleukin-2 (IL-2) cytokine prodrug engineered to preferentially activate effector CD8+ T cells and natural killer cells with reduced stimulation of regulatory T cells, and nivolumab, a monoclonal antibody targeting programmed cell death protein 1 (PD-1), blocking its interaction with ligands PD-L1 and PD-L2. The combination aimed to enhance antitumor immune responses by both promoting cytotoxic lymphocyte activation (via IL-2 agonism) and releasing inhibition of T-cell activity (via PD-1 blockade). Several large studies in advanced melanoma, renal cell carcinoma, bladder cancer, and sarcoma found no added clinical benefit over nivolumab monotherapy, and the global development program was discontinued due to a lack of efficacy gains and a higher rate of adverse events[1][2][4][5][6][7].
