Overview
**Bempegaldesleukin + nivolumab + axitinib** is an investigational combination regimen for advanced or metastatic renal cell carcinoma (RCC). - **Bempegaldesleukin** is a PEGylated recombinant interleukin-2 (IL-2) cytokine prodrug engineered to preferentially stimulate the IL-2 pathway via the CD122 (IL-2 receptor βγ) complex, thereby activating and expanding CD8^+ T cells and natural killer (NK) cells, with minimal expansion of immunosuppressive regulatory T cells. - **Nivolumab** is a monoclonal antibody checkpoint inhibitor targeting the programmed cell death 1 (PD-1) receptor, blocking PD-1/PD-L1 interaction and boosting T cell–mediated immune responses. - **Axitinib** is a small-molecule inhibitor of vascular endothelial growth factor receptors (VEGFRs), acting as an antiangiogenic targeted therapy. This triple combination is studied for its synergistic potential to enhance antitumor immunity and inhibit tumor angiogenesis, primarily in renal cell carcinoma.
