Overview

Bendamustine + dexamethasone + ixazomib (also referred to as BID or IBd) is an investigational triplet combination therapy primarily evaluated for the treatment of relapsed and/or refractory multiple myeloma (RRMM). The regimen combines three small molecule agents with distinct mechanisms: bendamustine, a bifunctional alkylating agent that causes DNA cross-linking; ixazomib, an orally bioavailable second-generation proteasome inhibitor that reversibly inhibits the β5 subunit of the 20S proteasome; and dexamethasone, a potent glucocorticoid that provides anti-inflammatory and pro-apoptotic effects. This combination has demonstrated clinical activity in heavily pretreated RRMM patients, including those previously exposed to bortezomib and lenalidomide, and has also shown potential antitumoral activity in preclinical models and case reports of hormone receptor-positive breast cancer.
