Overview

BGD is a combination chemotherapy regimen consisting of bendamustine, gemcitabine, and dexamethasone, specifically evaluated by the Polish Lymphoma Research Group (PLRG) for the treatment of relapsed or refractory classical Hodgkin lymphoma (cHL). This regimen serves as a salvage therapy to induce remission before autologous stem cell transplantation (ASCT) in patients who have failed first-line treatments like ABVD (adriamycin, bleomycin, vinblastine, dacarbazine). Gemcitabine is a nucleoside analog that inhibits DNA synthesis; bendamustine is a nitrogen mustard derivative that acts as a bifunctional alkylating agent; and dexamethasone is a corticosteroid that provides anti-inflammatory effects and modulates the immune response. Phase II studies, such as the BURGUND trial (PLRG-HL1), have demonstrated high complete metabolic response rates (67-84%) with this combination, showing it to be an effective and well-tolerated bridge to transplant.
