Berberine + paclitaxel is an experimental combination therapy involving the isoquinoline alkaloid berberine and the taxane microtubule stabilizer paclitaxel. Research has focused on enhancing the efficacy of this combination using advanced drug delivery systems, such as AI-guided CXCR4-targeted core-shell nanocarriers. These carriers typically consist of a mesoporous silica nanoparticle core for drug loading and a liposomal shell for biocompatibility and targeting. The combination aims to leverage the synergistic anti-tumor effects of both agents: paclitaxel induces mitotic arrest by stabilizing microtubules, while berberine inhibits cancer cell proliferation and migration, potentially reversing tumor-associated immune suppression (e.g., splenomegaly). The CXCR4-targeted formulation is specifically designed to improve selective uptake in CXCR4-positive cancer cells, such as those found in colorectal cancer, while reducing systemic toxicity.
