A synergistic combination of berberine (BBR) and wogonin (WOG) at an optimal ratio of 3:1, identified as the active components from the traditional Chinese medicine herb pair Coptidis Rhizoma and Scutellariae Radix. This research compound is designed to treat obesity-associated adipose tissue inflammation and insulin resistance by dually targeting the hypoxia-inducible factor alpha (HIF-α) axis. Specifically, it inhibits the pro-inflammatory Hypoxia-inducible factor 1-alpha (HIF-1α) while simultaneously activating Hypoxia-inducible factor 2-alpha (HIF-2α), which exerts anti-inflammatory effects. Preclinical studies in macrophage and adipocyte models, as well as high-fat diet-induced obese mouse models, have demonstrated its efficacy in improving insulin sensitivity and reducing metabolic inflammation.
