Drug intelligence / Profile preview
berobenatide + PF-08653945 is an investigational once-monthly subcutaneous combination therapy being developed by Pfizer for chronic weight management (obesity and overweight). The combination consists of berobenatide (PF-08653944, formerly MET-097i), an ultra-long-acting, fully biased glucagon-like peptide-1 receptor agonist (GLP-1 RA), and PF-08653945 (formerly MET-233i), an ultra-long-acting dual amylin and calcitonin receptor agonist (DACRA). Both components were originally developed by Metsera (acquired by Pfizer in late 2025) using its proprietary peptide stabilization and lipidation platform to achieve matched pharmacokinetic profiles suitable for once-monthly dosing. By combining GLP-1 receptor activation with amylin and calcitonin receptor agonism, the therapy targets complementary appetite-regulation and satiety pathways in the brain to enhance weight loss efficacy compared to monotherapy. The combination is currently being evaluated in the Phase 2b SOLIS-1 clinical trial.
