Overview

beta-caryophyllene + capsaicin is a combination of two naturally derived compounds, both with distinct mechanisms relevant to pain and inflammation. Beta-caryophyllene is a bicyclic sesquiterpene found in the essential oils of many plants including Cannabis sativa and functions as a selective agonist of the cannabinoid receptor 2 (CB2). It mediates anti-inflammatory and analgesic effects via CB2 receptor activation and secondary release of endogenous β-endorphin, affecting opioid signaling through peripheral mechanisms. Capsaicin is an active component of chili peppers and works as an agonist at the transient receptor potential vanilloid 1 (TRPV1), a non-selective cation channel in nociceptive sensory neurons, leading to desensitization, local substance P depletion, and analgesia upon repeated application. The combination of beta-caryophyllene and capsaicin has been studied for topical use, particularly in chronic pain management, leveraging the distinct but potentially complementary analgesic mechanisms of both compounds[1][4]. While beta-caryophyllene may increase analgesic efficacy and modulate capsaicin-induced nociception, the combination is still under evaluation and is not widely marketed as a pharmaceutical product.
