Betamethasone + 5-fluorouracil is a combination therapy consisting of the synthetic glucocorticoid betamethasone and the antimetabolite 5-fluorouracil. It is primarily used as an intralesional injection for the treatment of keloids and hypertrophic scars. Betamethasone acts by suppressing inflammation and inhibiting collagen synthesis through glucocorticoid receptor activation, while 5-fluorouracil inhibits fibroblast proliferation and TGF-beta-induced collagen expression by interfering with DNA synthesis via thymidylate synthase inhibition. This synergistic approach helps reduce the volume and recurrence rate of keloids, which are often resistant to monotherapy.
