Celestone Chronodose is an injectable corticosteroid formulation composed of a mixture of soluble betamethasone sodium phosphate and a less soluble betamethasone acetate suspension. This combination provides a dual-action profile: the sodium phosphate component offers rapid anti-inflammatory activity, while the acetate component provides sustained release (depot effect) over several days. In the clinical context of ophthalmic surgery, specifically cataract extraction, it is administered as a single subconjunctival injection to prevent postoperative intraocular inflammation and macular edema. The drug acts as a potent glucocorticoid receptor agonist, suppressing the production of inflammatory mediators and reducing vascular permeability. It has been investigated by Oogziekenhuis Rotterdam as an alternative to traditional topical steroid eye drop regimens following cataract surgery.
