Overview

This regimen is a **combination therapy** consisting of four drugs: - **Bevacizumab**: a humanized monoclonal antibody targeting vascular endothelial growth factor (VEGF), thereby inhibiting angiogenesis. - **5-fluorouracil**: an antimetabolite and pyrimidine analog that is incorporated into RNA and DNA, leading to inhibition of thymidylate synthase and cell death. - **Leucovorin calcium**: a reduced folate used to enhance the cytotoxic effect of 5-fluorouracil. - **Oxaliplatin**: a platinum-based chemotherapeutic agent that exerts its effect by forming DNA crosslinks. This combination, commonly referred to as **FOLFOX plus bevacizumab**, is widely used as a first-line chemotherapeutic regimen for metastatic colorectal cancer. The combination acts through multiple mechanisms: inhibition of tumor angiogenesis (bevacizumab), interference with DNA synthesis and repair (5-fluorouracil and oxaliplatin), and rescue/potentiation of 5-fluorouracil cytotoxicity (leucovorin)[1][2][3].
