Overview

This is a four-drug combination regimen consisting of bevacizumab, carboplatin, cixutumumab, and paclitaxel. Bevacizumab is a monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), thereby blocking angiogenesis in tumors. Carboplatin is a platinum-based chemotherapeutic agent that causes DNA crosslinking and apoptosis in rapidly dividing cells. Paclitaxel is a microtubule-stabilizing agent that disrupts mitosis by preventing microtubule depolymerization. Cixutumumab (IMC-A12) is a monoclonal antibody targeting the insulin-like growth factor 1 receptor (IGF-1R), inhibiting IGF-mediated signaling pathways involved in tumor cell proliferation and survival. This specific four-drug combination has been studied primarily in advanced or metastatic solid tumors such as non-small cell lung cancer, with the rationale of combining cytotoxic chemotherapy with antiangiogenic therapy and targeted inhibition of IGF signaling[6][8]. Each component acts via distinct mechanisms to inhibit tumor growth.
