The combination of bevacizumab and cholecalciferol (vitamin D3) is currently being studied in clinical trials for treating metastatic colorectal cancer. Bevacizumab is a recombinant, humanized anti-vascular endothelial growth factor (VEGF) monoclonal antibody that neutralizes VEGF's biological activity and inhibits tumor angiogenesis. Cholecalciferol (vitamin D3) is being investigated as a potential addition to standard chemotherapy regimens that include bevacizumab. Clinical trials, including a phase III trial, are examining whether giving vitamin D3 with chemotherapy and bevacizumab works better in shrinking or stabilizing colorectal cancer compared to chemotherapy and bevacizumab alone. A phase II trial is also studying high-dose cholecalciferol in treating patients receiving combination chemotherapy and bevacizumab as first-line therapy for metastatic colorectal cancer, investigating if cholecalciferol may delay the development of colorectal cancer. The combination therapy typically includes bevacizumab along with standard chemotherapy agents such as leucovorin calcium, fluorouracil, oxaliplatin, and sometimes irinotecan hydrochloride, plus high-dose vitamin D3 (cholecalciferol).
