bevacizumab + dacarbazine + interferon alfa-2a is a combination therapy regimen investigated as a first-line treatment for patients with locally advanced or metastatic melanoma. This therapeutic approach, evaluated in a Phase 2 study (NCT00779714) led by the University of Turku, combines the anti-angiogenic monoclonal antibody bevacizumab (Avastin), the alkylating cytotoxic agent dacarbazine, and the immunomodulatory cytokine interferon alfa-2a (Roferon-A). Bevacizumab works by binding to vascular endothelial growth factor (VEGF), thereby inhibiting tumor angiogenesis. Dacarbazine acts as a chemotherapy agent that methylates DNA to induce cell death. Interferon alfa-2a enhances the host immune response and exerts direct anti-proliferative effects on tumor cells. The study aimed to assess whether this triple combination could improve efficacy over standard treatments in the advanced melanoma setting.
