Overview

This combination therapy is a Phase II immunotherapy regimen developed and studied at Dartmouth-Hitchcock Medical Center for the treatment of metastatic renal cell carcinoma (mRCC). The regimen integrates four distinct therapeutic components: bevacizumab (a monoclonal antibody targeting VEGF), an autologous dendritic cell (DC) vaccine loaded with tumor lysate, interleukin-2 (aldesleukin), and interferon alfa-2b. The approach aims to combine immune activation with the disruption of tumor-induced immunosuppression and angiogenesis. Interleukin-2 (administered as a continuous intravenous infusion) and interferon alfa-2b act as cytokines to promote T-cell and NK-cell proliferation and effector function, while the DC vaccine provides specific tumor-antigen presentation to prime the immune system. Bevacizumab is included to block vascular endothelial growth factor (VEGF), which is often elevated in mRCC and associated with poor response to cytokine therapy and systemic immune dysregulation.
