Overview

Bevacizumab + erlotinib + panitumumab is a triple-drug combination therapy investigated for the treatment of non-small-cell lung cancer (NSCLC). This regimen combines three distinct targeted agents: bevacizumab, a monoclonal antibody that inhibits vascular endothelial growth factor (VEGF) to prevent angiogenesis; erlotinib, a small molecule tyrosine kinase inhibitor (TKI) targeting the epidermal growth factor receptor (EGFR); and panitumumab, a monoclonal antibody that also targets EGFR by binding to its extracellular domain. A Phase III clinical trial conducted in Chinese patients with progressed NSCLC after first-line platinum-based chemotherapy demonstrated that this combination as a second-line therapy significantly improved progression-free survival (4.6 months vs 1.9 months) and overall survival (10.4 months vs 8.9 months) compared to erlotinib monotherapy. However, clinical use has been limited by concerns regarding increased severe adverse events associated with combining multiple VEGF and EGFR inhibitors.
