Overview

Bevacizumab + everolimus + panitumumab (BEP) is an investigational triple-drug combination regimen evaluated for the treatment of refractory advanced solid tumors. This combination therapy simultaneously targets three distinct oncogenic pathways: angiogenesis via bevacizumab (a monoclonal antibody targeting vascular endothelial growth factor, VEGF), the mTOR pathway via everolimus (a small molecule mTOR inhibitor), and the epidermal growth factor receptor (EGFR) pathway via panitumumab (a fully human monoclonal antibody targeting EGFR). The rationale for this triplet is to achieve synergistic antitumor and antiangiogenic effects by concurrently inhibiting these interconnected signaling pathways. A Phase I study led by Duke University established the safety profile and recommended Phase II dosing for this combination, observing prolonged clinical activity in several tumor types.
