Overview

Bevacizumab + FOLFIRI-3 is a combination chemotherapy and targeted therapy regimen primarily used in the treatment of metastatic colorectal cancer, especially after failure of standard first-line therapies. The regimen consists of three chemotherapeutic agents—fluorouracil (5-FU), leucovorin calcium (folinic acid), and irinotecan hydrochloride—combined with the monoclonal antibody bevacizumab. Fluorouracil is a pyrimidine analog that inhibits DNA synthesis; leucovorin enhances its efficacy; irinotecan is a topoisomerase I inhibitor that prevents DNA replication; and bevacizumab targets vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis required for tumor growth. This combination has shown activity in patients with metastatic colorectal cancer who have progressed on previous lines of chemotherapy[1][3][5].
