Overview

Bevacizumab + onartuzumab is an investigational combination therapy consisting of two monoclonal antibodies: bevacizumab, a humanized antibody targeting vascular endothelial growth factor A (VEGF-A), and onartuzumab, a monovalent antibody that targets the hepatocyte growth factor receptor MET. Bevacizumab inhibits angiogenesis by blocking VEGF-A, thereby reducing tumor blood supply. Onartuzumab blocks MET signaling, which is implicated in tumor cell proliferation, survival, invasion, and resistance to antiangiogenic therapies. The combination has been studied in various solid tumors including non-small cell lung cancer (NSCLC) and metastatic colorectal cancer (mCRC), aiming to overcome resistance mechanisms associated with VEGF inhibition by simultaneously targeting the MET pathway. Clinical trials have shown that while the regimen is generally well tolerated with manageable side effects such as fatigue and peripheral edema, it did not demonstrate significant improvement in efficacy outcomes compared to standard regimens[1][2][5][6].
