Overview

This is a combination therapy that has been studied in clinical trials, particularly for triple-negative breast cancer (TNBC) and non-small cell lung cancer (NSCLC). The combination consists of three distinct drugs: 1. Bevacizumab (Avastin) - an anti-angiogenic monoclonal antibody targeting VEGF 2. Onartuzumab (MetMAb) - a monovalent monoclonal antibody targeting the MET receptor 3. Paclitaxel - a cytotoxic chemotherapy agent Clinical trials (e.g., OAM4861g for TNBC, NCT01496742 for NSCLC) evaluating this combination did not demonstrate improved efficacy with the addition of onartuzumab. In fact, for TNBC, progression-free survival was worse in the onartuzumab-containing arms compared to bevacizumab plus paclitaxel alone. Similar lack of benefit and shorter overall survival were observed in other studies. The combination was associated with adverse events such as peripheral edema and increased venous thromboembolic events.
