This is a combination regimen consisting of three agents: - **Bevacizumab** is a monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis and thereby restricting tumor blood supply. - **Oxaliplatin** is a platinum-based small molecule chemotherapeutic that induces DNA crosslinking, leading to apoptosis in rapidly dividing cells. - **Panitumumab** is a fully human monoclonal antibody targeting the epidermal growth factor receptor (EGFR), blocking EGFR-mediated signaling pathways involved in cell proliferation and survival. This combination has been studied as first-line treatment for metastatic colorectal cancer (mCRC). However, clinical trials have shown that adding panitumumab to bevacizumab and oxaliplatin-based chemotherapy results in increased toxicity—including higher rates of skin toxicity, diarrhea, infections, pulmonary embolism—and decreased progression-free survival compared to regimens without panitumumab. No subset of patients with wild-type or mutant KRAS tumors benefited from this triple combination. As such, this regimen is not recommended for mCRC treatment in clinical practice[3][5][1].
