Bevacizumab + pemetrexed is a combination therapy used primarily in the treatment of advanced nonsquamous non-small-cell lung cancer (NSCLC) and has also shown efficacy in recurrent and metastatic cervical cancer. Bevacizumab is a recombinant humanized monoclonal antibody that inhibits angiogenesis by binding to vascular endothelial growth factor A (VEGF-A), thereby blocking the formation of new blood vessels necessary for tumor growth. Pemetrexed is a small molecule antimetabolite that disrupts folate-dependent metabolic processes essential for cell replication, inhibiting enzymes involved in purine and pyrimidine synthesis. The combination leverages both antiangiogenic and cytotoxic mechanisms to improve clinical outcomes, including progression-free survival, compared to monotherapy with either agent[1][2][3][5].
