This combination therapy is a multi-agent regimen consisting of the angiogenesis inhibitor bevacizumab, the folate antimetabolite pemetrexed, and a platinum-based chemotherapeutic agent (either cisplatin or nedaplatin). It is primarily being investigated as a first-line treatment for patients with EGFR/ALK-negative non-squamous non-small cell lung cancer (NSCLC) who present with brain metastases. Bevacizumab is a monoclonal antibody that targets vascular endothelial growth factor (VEGF), inhibiting tumor angiogenesis and potentially enhancing the penetration of chemotherapy across the blood-brain barrier. Pemetrexed disrupts folate-dependent metabolic processes essential for DNA synthesis, while platinum agents induce DNA cross-linking, collectively leading to tumor cell apoptosis. The regimen is currently being evaluated in a Phase 3 clinical trial sponsored by Sun Yat-sen University.
