Overview

Combination of two monoclonal antibody biologics used investigationally together to treat cancer. Bevacizumab is an anti-angiogenic antibody that binds and neutralizes VEGF-A to inhibit tumor angiogenesis, while pertuzumab is a HER2 dimerization inhibitor that binds HER2 subdomain II to block ligand-dependent HER2 heterodimerization (with EGFR/HER1, HER3, HER4), suppress downstream signaling (MAPK/PI3K), and enhance antibody-dependent cell-mediated cytotoxicity. The combination has been explored in oncology settings alongside or compared with other HER2-directed regimens; it is not an approved fixed-dose product as a pair and is distinct from the approved pertuzumab+trastuzumab combinations (e.g., IV co-administration and the subcutaneous fixed-dose with hyaluronidase). Bevacizumab was first approved in 2004 as Avastin for multiple cancers, and pertuzumab (Perjeta) was first approved in 2012 for HER2-positive breast cancer in combination with trastuzumab and docetaxel.[5][1][2][4][6][9]
