Overview

Bevacizumab + rindopepimut is a combination therapy investigated primarily for the treatment of recurrent glioblastoma that expresses the EGFRvIII mutation. Bevacizumab is a monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A), thereby inhibiting angiogenesis and reducing tumor blood supply. Rindopepimut (CDX-110) is an investigational peptide vaccine that targets the tumor-specific epidermal growth factor receptor variant III (EGFRvIII), stimulating an immune response against cells expressing this mutant protein. The rationale for combining these agents lies in their complementary mechanisms—bevacizumab may reduce VEGF-mediated immunosuppression in the tumor microenvironment, potentially enhancing the efficacy of immune-based therapies like rindopepimut. Clinical studies, including the phase II ReACT trial, have shown that this combination can improve overall survival and progression-free survival compared to bevacizumab alone in patients with relapsed EGFRvIII-positive glioblastoma[1][3][9].
