bevacizumab + sapanisertib is a combination therapy investigated for the treatment of recurrent glioblastoma and other advanced solid tumors. It consists of bevacizumab, a recombinant humanized monoclonal antibody that targets vascular endothelial growth factor (VEGF) to inhibit tumor angiogenesis, and sapanisertib (formerly MLN0128), an orally bioavailable small molecule that acts as a potent and selective inhibitor of both mTORC1 and mTORC2. By simultaneously blocking VEGF-mediated blood vessel growth and the PI3K/AKT/mTOR intracellular signaling pathway, the combination aims to overcome resistance mechanisms and provide enhanced anti-tumor activity compared to monotherapy. This combination has been primarily evaluated in Phase 1 clinical trials sponsored by the National Cancer Institute.
