Overview

Bevacizumab + temozolomide is a combination therapy primarily used in the treatment of high-grade gliomas and glioblastoma. Bevacizumab is a monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), thereby blocking angiogenesis and reducing tumor blood supply. Temozolomide is an oral alkylating agent that induces DNA damage leading to apoptosis in tumor cells. The combination aims to both impair tumor angiogenesis (via bevacizumab) and directly kill cancer cells (via temozolomide). This regimen has been shown to improve progression-free survival and overall survival compared to monotherapy with temozolomide alone, though it may be associated with increased adverse events such as thrombocytopenia, neutropenia, leukopenia, anemia, and fatigue[1][2][4][5]. The combination does not typically result in cure but can prolong life and improve symptoms for patients with aggressive brain tumors.
