**AVANTHER** was a Phase 2, perioperative HER2-positive breast-cancer treatment regimen studied by Hospital Universitario Madrid Sanchinarro. It administered neoadjuvant bevacizumab, trastuzumab, and weekly paclitaxel before surgery, followed by adjuvant trastuzumab, cyclophosphamide, and non-pegylated liposomal doxorubicin supplied as Myocet. Bevacizumab neutralizes VEGF-A to suppress tumor angiogenesis; trastuzumab targets HER2; paclitaxel stabilizes microtubules; cyclophosphamide alkylates DNA after metabolic activation; and liposomal doxorubicin intercalates DNA and inhibits topoisomerase II.
