Overview

Bevacizumab + vismodegib is an investigational combination therapy consisting of two distinct agents with complementary mechanisms of action, primarily explored in oncology. Bevacizumab is a monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), thereby blocking angiogenesis and reducing blood supply to tumors. Vismodegib is a small molecule inhibitor targeting the Hedgehog signaling pathway by antagonizing Smoothened (SMO), which plays a critical role in tumor cell proliferation and survival, particularly in basal cell carcinoma and other cancers with aberrant Hedgehog pathway activation. The combination has been studied in clinical trials for metastatic colorectal cancer (mCRC) as an adjunct to standard chemotherapy regimens, but did not demonstrate added efficacy over standard therapy alone[1]. Both drugs are approved individually for various indications but are not approved as a fixed-dose combination.
