BG-C477 + chemotherapy is a combination therapy being investigated for the treatment of advanced, metastatic, or unresectable solid tumors. The combination consists of BG-C477, an antibody-drug conjugate (ADC) targeting carcinoembryonic antigen-related cell adhesion molecule 5 (CEACAM5, also known as CEA), and various systemic chemotherapy agents. BG-C477 is designed to selectively deliver a cytotoxic payload to tumor cells overexpressing CEACAM5, a protein frequently found in gastrointestinal and non-small cell lung cancers, while having limited expression on normal tissues. The addition of chemotherapy is intended to provide synergistic antitumor activity. This combination is currently being evaluated in a Phase 1a/b multicenter, open-label, first-in-human study (NCT06596473) to assess safety, tolerability, pharmacokinetics, and preliminary efficacy in patients who have progressed on at least two prior lines of standard systemic therapy.
