**BGB-10188 + tislelizumab** is an investigational combination therapy being studied in advanced solid tumors and B-cell malignancies. BGB-10188 is an orally available, highly selective inhibitor of phosphoinositide 3-kinase delta (PI3Kδ), which plays a key role in B-cell receptor signaling and regulatory T-cell (Treg) function. Inhibition of PI3Kδ can suppress Treg proliferation and enhance anti-tumor immunity by optimizing CD8+ T-cell activation. Tislelizumab is a monoclonal antibody targeting the programmed death-1 (PD-1) receptor, functioning as a checkpoint inhibitor to block PD-1/PD-L1 interactions, thereby enhancing T-cell–mediated anti-tumor immune responses. Preclinical and early clinical studies suggest the combination exhibits synergistic antitumor activity, with manageable safety and tolerability, particularly in heavily pretreated patients with solid tumors.
