Overview

BGB-24714 + docetaxel is a combination therapy under investigation for the treatment of advanced or metastatic solid tumors. BGB-24714 is a novel oral inhibitor of apoptosis protein (IAP) antagonist, specifically a second mitochondrial-derived activator of caspases (SMAC) mimetic. It functions by promoting the degradation of cellular IAP1 (cIAP1), antagonizing X-linked inhibitor of apoptosis protein (XIAP), and facilitating caspase-9 activation, thereby promoting apoptosis in tumor cells. Docetaxel is a well-established chemotherapeutic agent of the taxane class that stabilizes microtubules, inhibiting mitosis and promoting cancer cell death. The combination is being studied to determine its safety, tolerability, and efficacy, with preclinical models showing synergistic antitumor activity when BGB-24714 is combined with chemotherapy, especially docetaxel and paclitaxel[1][2][3][4][5][6][7].
