BGB-24714 + paclitaxel is a combination therapy under investigation for the treatment of advanced or metastatic solid tumors. BGB-24714 is a novel, orally administered small molecule antagonist of inhibitor of apoptosis proteins (IAPs), including cellular IAP1 (cIAP1) and X-linked IAP (XIAP). It acts as a SMAC mimetic, disrupting the inhibition of apoptosis in cancer cells. Paclitaxel is a well-established chemotherapeutic agent that stabilizes microtubules, thereby inhibiting cell division. Preclinical studies have shown this combination produces synergistic anti-tumor effects, particularly in breast cancer models, by enhancing apoptosis and tumor growth inhibition compared to either agent alone. The combination is currently in phase 1 clinical trials for safety, tolerability, and preliminary efficacy in patients with solid tumors[1][2][3][4][5][7].
