BI-1607 + trastuzumab is a combination therapy under clinical investigation for the treatment of HER2-positive advanced solid tumors, including breast and gastric cancers. BI-1607 is an engineered human IgG1 monoclonal antibody targeting CD32b (Fc gamma receptor IIB, FcyRIIB), designed to enhance the efficacy and overcome resistance to existing cancer treatments such as trastuzumab. Trastuzumab is a well-established monoclonal antibody that targets the human epidermal growth factor receptor 2 (HER2). The rationale for this combination is that many patients with HER2+ cancers develop resistance to trastuzumab; by inhibiting FcyRIIB, BI-1607 may potentiate immune-mediated anti-tumor activity and improve response rates in these patients. Early clinical data indicate that the combination has been well tolerated with stable disease observed in several patients who had previously progressed on trastuzumab[2][5][6][8].
