BI 1703880 + ezabenlimab is an investigational combination therapy being evaluated for advanced solid tumors. BI 1703880 is a novel STimulator of INterferon Genes (STING) agonist that promotes innate immune activation and upregulates programmed death ligand 1 (PD-L1) and human leukocyte antigen on tumor cells. Ezabenlimab is an anti-programmed cell death protein 1 (PD-1) monoclonal antibody checkpoint inhibitor that blocks PD-1 on T cells, thus enhancing adaptive immune responses against cancer. The rationale for combining these agents is that STING pathway activation may sensitize tumors to PD-1 inhibition, potentially overcoming resistance to immunotherapy. This combination is under investigation in an open-label, phase Ia dose-escalation clinical trial (NCT05471856) in adults with advanced metastatic or recurrent solid malignancies who have exhausted standard therapies[1][2][3][5][7][8].
