Overview

BI 1823911 + BI 1701963 is an investigational combination therapy for solid tumors with a KRAS G12C mutation. BI 1823911 is a highly potent, irreversible inhibitor targeting KRAS G12C in its GDP-bound state, while BI 1701963 is a pan-KRAS SOS1 inhibitor designed to shift KRAS G12C towards its GDP-loaded form, enhancing the binding and efficacy of BI 1823911. The combination shows increased pathway modulation and deeper anti-tumor efficacy than monotherapy, with synergistic effects observed in preclinical and early clinical studies. Developed primarily for advanced/metastatic solid tumors that are therapy-naïve or have relapsed, including non-small cell lung cancer (NSCLC), colorectal cancer (CRC), cholangiocarcinoma, and pancreatic adenocarcinoma. The drug is currently in Phase 1 clinical trials to evaluate its safety, pharmacokinetics, and preliminary efficacy[1][2][3][4].
