**BI 1831169 + nivolumab** is an investigational combination therapy under development for advanced and metastatic solid tumors. BI 1831169, also known as VSV-GP, is a recombinant oncolytic virus based on vesicular stomatitis virus in which the native glycoprotein G is replaced by that of the lymphocytic choriomeningitis virus (LCMV). This results in a live, replication-competent viral immunotherapy that selectively targets and lyses tumor cells (oncolysis), while also stimulating anti-tumor immune responses. Nivolumab is a programmed death-1 (PD-1) immune checkpoint inhibitor already approved for multiple cancers; in this combination, it is intended to further enhance antitumor immune activity by blocking the PD-1 pathway, lifting the braking mechanism that tumors use to suppress immune attack. The duo is administered intravenously and aims to maximize tumor-specific immune activation by combining direct oncolytic activity with immune checkpoint blockade.
