BI 2536 + gemcitabine is a combination therapy studied primarily for the treatment of locally advanced or metastatic pancreatic cancer. BI 2536 is a highly selective, ATP-competitive, small molecule inhibitor of the Polo-like kinase family (primarily PLK1, also PLK2 and PLK3), key regulators of mitosis and cell cycle progression. By inhibiting PLK1, BI 2536 induces mitotic arrest and apoptosis in tumor cells. Gemcitabine is an antineoplastic nucleoside analog that inhibits DNA synthesis, commonly used as standard chemotherapy in pancreatic cancer. When co-administered, BI 2536 has shown to sensitize gemcitabine-resistant pancreatic cancer cells and tumors to gemcitabine treatment, demonstrating synergistic anti-tumor activity in preclinical and clinical studies. Combination therapy aims to improve response rates compared to gemcitabine monotherapy by overcoming chemoresistance via multi-targeted mechanisms[1][3][7].
